Description:
Cytokines of the IL6/GCSF/MGF family are glycoproteins of about 170 to 180 amino acid residues that contains four conserved cysteine residues involved in two disulphide bonds. They have a compact, globular fold (similar to other interleukins), stabilised by the 2 disulphide bonds. One half of the structure is dominated by a 4 alpha-helix bundle with a left-handed twist: the helices are anti-parallel, with 2 overhand connections, which fall into a 2- stranded anti-parallel beta-sheet. The fourth alpha-helix is important to the biological activity of the molecule. Interleukin (IL)-6 is an important proinflammatory and immunoregulatory cytokine expressed by various cells.Interleukin-6 has been shown to inhibit the growth of early stage and to promote the proliferation of advanced stage melanoma cells in vitro. Recombinant Human IL-6 produced in E.Coli is a single, non-glycosylated polypeptide chain containing 184 amino acids and having a molecular mass of 21,000 Daltons.

Formulation:
Lyophilized from 10 mM Acetic acid.
Reconstitution:
Centrifuge vial before opening. When reconstituting the product, gently pipet and wash down the sides of the vial to ensure full recovery of the protein into solution. It is recommended to reconstitute the lyophilized product with 10 mM HCl at a concentration of 0.1 mg’s/ml, which can be further diluted into other aqueous solutions.
Stability:
Lyophilized product is very stable at -20 degrees. Reconstituted material should be aliquoted and frozen at -20. It is recommended to add a carrier protein (0.1% HSA or BSA) for long term storage.
Purity Greater than 95% as determined by:
Analysis by RP-HPLC.
Reducing and non-reducing SDS-PAGE.
Protein Content determined by:
UV spectroscopy at 280 nm.
Analysis by RP-HPLC calibrated against a known standard.
Quantitation on SDS-PAGE against a known standard.
Endotoxin Level:
Endotoxin level as measured by LAL is <0.01 ng/ug or <0.1EU/ug.
Biological Activity:
Human IL-6 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of mouse 7TD1 cells is less then 0.08 ng/ml.
Stem Cell Related Validation:
Recombinant Interleukin 6 was shown to have excellent activity for the induction of human fibroblasts for use as feeder cells
