Exosome Marker Antibodies for EV Characterisation

Robust characterisation of extracellular vesicles (EVs) is a requirement of any publishable study. Community guidance on the minimal information for EV studies (MISEV) recommends demonstrating the presence of at least one tetraspanin marker alongside a non-vesicle protein as a negative control. Cell Guidance Systems offers a validated suite of reagents to meet these requirements, including exosome marker antibodies, the ExoLISA™ detection assay, and an NTA size profiling service.

Validated CD9, CD63 and CD81 Antibodies

These monoclonal antibodies target the three most widely used exosome tetraspanin markers: CD9, CD63, and CD81. Each antibody has been validated against human and murine exosome-associated antigens and specifically tested for western blotting of EV preparations from cell culture conditioned media and biological fluids. All clones are produced in-house and quality-controlled for lot-to-lot consistency.

Why use tetraspanin antibodies for EV characterisation?

CD9, CD63, and CD81 are integral membrane proteins highly enriched on the surface of exosomes and small EVs. They are accepted as positive EV markers by the MISEV guidelines and are routinely detected by western blot. Using all three markers together (available as the EX205 panel bundle) provides stronger evidence of vesicle identity than a single marker alone.
