Description
Activin A is a critical component of directed differentiation protocols for iPSC and embryonic stem cell lines. It is most widely used for definitive endoderm induction, where it acts through Nodal/Activin signalling to drive mesendoderm and endoderm fate specification. Standard protocols combine Activin A with Wnt pathway activators (such as CHIR99021) over 3–5 days to generate CXCR4+/SOX17+ definitive endoderm cells, as a precursor to pancreatic, hepatic, and intestinal lineage differentiation. Activin A is also used to maintain pluripotency in human and mouse ESC culture, and as a TGF-beta superfamily ligand in studies of wound healing, reproductive biology, and haematopoiesis.
Activity Assay
Human Activin A is fully biologically active when compared to standard. The activity is determined using MPC-11 cells and it is typically 1.6 - 3.1 ng/ml. In addition to the standard assay, the activity of this product has been verified in the lab of Dr Josué Mfopou Kunjom (VUB - Diabetes Research Centre) - by analysis of CXCR4 in definitive endoderm.
